CD68 is a highly glycosylated transmembrane protein, which is the most widely used macrophage marker. CD68 is expressed in Kupffer cells, normal lymphoid cells, mast cells and microglia. In tumor tissue, CD68 is expressed in the epithelial cells of fibrous tissue tumors, some epithelial tumors, and some malignant melanoma.
CD74 is a membrane binding subunit of the class II related invariant chain of the major histocompatibility complex. CD74 is mainly used to label lymphocytes in germinal center and in the mantle cell area of lymph nodes. This does not label plasma cells or thymocytes, but usually labels dendritic cells. About 90% of low-grade B-cell lymphoma and 20% of low-grade T-cell lymphoma are expressing CD74, but in high-grade lymphoma, the ability to distinguish B cells from T cells is decreased.
