GST can catalyze the binding of reduced glutathione (GSH) to dinitrobenzene (CDNB) and the product have an absorption peak at 340 nm. The activity of GSH-ST can be calculated by measuring the increasing rate of absorbance at 340 nm.
Glutathione S-transferase is a kind of enzyme related to liver detoxification, which is often used as an indicator of liver injury. GST can resist the damage of endogenous and exogenous electronophilic substances, and plays an important role in the anti-tumor process.
Glyceraldehyde-3-Phosphate Dehydrogenase (GAPDH), a key enzyme in the glycolytic pathway, catalyzes the oxidation of glyceraldehyde-3-phosphate to 1, 3-diphosphoglycerate. This response is closely related to the gluconeogenic pathway, the maintenance of blood glucose concentrations in the body, and the development of diabetes. GAPDH gene is a housekeeping gene, which is expressed at a high level in almost all tissues. Therefore, GAPDH widely exists in tissues and cells and plays an important role in the disorders of glucose, lipid and protein metabolism in the body. This kit can calculate the enzyme activity of GAPDH by measuring the OD value change at 450 nm per unit time of the product produced by GAPDH catalyzing the oxidation of glyceraldehyde-3-phosphate.
Glycerol 3-phosphate (G3P), also known as 3-glycerophosphoic acid and glycerol α-phosphate, is mainly derived from glycolysis. G3P is an important intermediate in the glycerol-3-phosphate shuttle mechanism and studies have shown that G3P is involved in electron transport, and its metabolic level is related to lipid metabolism, insulin resistance and some cancers. The principle of this kit is that G3P is decomposed by enzymes, and its products generate fluorescent substances with probes under the action of other enzymes. The higher the fluorescence value, the higher the G3P content in the sample.
Glycerol is transformed by enzyme to produce hydrogen peroxide, which is catalyzed by oxidase in the presence of 4-amino-antipyrprid and phenol to produce red quinones, the color of which is proportional to the content of glycerol.
Glycerol kinase (GK) is a rate-limiting enzyme in glycerol metabolism, and the lack of this enzyme directly results in the inability of cells to utilize glycerol. This kit produces hydrogen peroxide under the action of glycerophosphate oxidase. The reaction between hydrogen peroxide and the chromogenic agent has the maximum absorption peak at 555 nm. The enzyme activity of GK can be determined by measuring the increase rate at 555 nm.
Under the presence of concentrated sulfuric acid, glycogen can be dehydrated to furfural derivatives. Furfural derivatives can form blue compound with anthracenone. The concentration of the compound can be measured by colorimetric quantification at 620 nm with glucose standard buffer of same treatment. Glycogen is quite stable in concentrated alkali solution. Heating the tissue sample in concentrated alkali solution before color development will remove other components and keep the glycogen.
