Alpha-glucosidase catalyzes the release of α-glucose from the non-reducing end of the substrate and promotes the absorption of glucose by the small intestine. Inhibition of α-glucosidase is an effective method for the treatment of non-insulin-dependent diabetes mellitus (NIDDM). The detection principle of this kit: α-Glucosidase catalyzes the substrate reaction to generate colored products, whose absorbance increases at 400 nm. The addition of an inhibitor suppresses the enzymatic activity of α-glucosidase, resulting in a reduced rate of absorbance increase. The inhibition rate can be calculated based on the absorbance difference.
Ammonia is an important source of nitrogen for living systems and is necessary for the synthesis of amino acids. Ammonia is a metabolite produced by the deamination of amino acids. It plays an important role in both normal and abnormal animal physiology. In severe liver diseases, ammonia can not be removed from the circulation, resulting in increased blood ammonia, which can lead to hepatic encephalopathy (hepatic coma). In clinical practice, blood ammonia measurement is an important experimental diagnosis and monitoring index of hepatic encephalopathy. The detection principle of this kit: enzyme-catalyzed ammonia reaction consumes NADH, causing its absorbance to decline at 340 nm. By measuring the rate of absorbance decline at 340 nm, the ammonia content can be measured.
Amylopectin can be combined with the chromogenic component at 530 nm for maximum absorption, and the dual-wavelength method can be used to determine the content of amylose more accurately.
Amylose can be combined with the chromogenic component at 600 nm for maximum absorption, and the dual-wavelength method can be used to determine the content of amylose more accurately.
Angiotensin Converting Enzyme 1(ACE1), also known as angiotensin converting enzyme (ACE), kininase II, and peptidy-carboxylpeptidase, is an important component of the renin-angiotensin system (RAS). In the RAS system, angiotensin is converted into angiotensin II. Angiotensin II can cause strong contractions of vascular smooth muscle, increasing peripheral vascular resistance and thereby leading to elevated blood pressure. Excessively high ACE1 activity can directly result in elevated blood pressure. ACE1 inhibitors can achieve the purpose of lowering blood pressure by inhibiting ACE1 activity. The detection principle of this kit: ACE1 catalyzes the decomposition of the substrate, releasing fluorescent products. After adding the inhibitor, the generation of fluorescent substances will be inhibited, and the effect of the inhibitor is determined based on the degree of inhibition.
N-(3-(2-furyl) acryloyl)-L-phenylalanyl-glycyl-glycine (FAPGG) have the maximum absorption peak at 340 nm. ACE1 catalyzes hydrolysis of FAPGG substrate to furanyl phenylalanine (FAP) and glycylglycine (GG), and the absorbance at 340 nm will be decreased. The activity of ACE1 can be calculated indirectly by measuring the decrease in absorbance at 340 nm.
