B2M, also known as β2-Microglobulin or CDABP0092, is a component of MHC class I molecules found expression in all nucleated cells (excludes red blood cells). The major function of MHC class I moleculesis is to display fragments of proteins from within the cell to T-cells and cells containing foreign proteins will be attacked. B2M (β2-Microglobulin) is a low molecular weight protein. It was demonstrated that B2M (β2-Microglobulin) was localized in the membranes of nucleated cells and was found to be associated with HL-A antigens. B2M (β2-Microglobulin) is present in free form in various body fluids and as a subunit of histocompatibility antigens on cell surfaces lateral to the α3 chain. Unlike α3, β2 has no transmembrane region. Directly above β2 lies the α1 chain, which itself is lateral to the α2. In the absence of B2M (β2 microglobulin), very limited amounts of MHC class I (classical and non-classical) molecules can be detected on the surface. In the absence of MHC class I, CD8 T cells, a subset of T cells involved in the development of acquired immunity cannot develop. Low levels of B2M (β2 microglobulin) can indicate non-progression of HIV.
β-2-Microglobulin (B2M) is a secreted protein with 1 Ig-like C1-type (immunoglobulin-like) domain which belongs to the beta-2-microglobulin family. B2M component of major histocompatibility complex (MHC) class I molecules, involved in the presentation of peptide antigens to the immune system. Polymers of beta 2-microglobulin can be found in tissues from patients on long-term hemodialysis. B2M is a protein found on the surface of many cells and plentiful on the surface of white blood cells. Serum B2M concentration is increased in renal diseases, various malignant diseases and some inflammatory and autoimmune disorders. B2M may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. B2M has been shown as a marker for monitoring inflammatory disease activity and it appears likely to have a destructive role in amyloidosis-related arthritis. B2M might be involved in the OA (osteoarthritis) pathogenesis. Defects in B2M are the cause of hypercatabolic hypoproteinemia. Affected individuals show marked reduction in serum concentrations of immunoglobulin and albumin, probably due to rapid degradation. B2M could be a potential therapeutic target in ovarian cancer.
Programmed death ligand 2 (PD-L2), also referred to as B7-DC and CD273, is a member of the B7 family of proteins including B7-1, B7-2, B7-H2, B7-H1 (PD-L1), and B7-H3. PD-L2 is a type I membrane protein and structurally consists of an extracellular region containing one V-like and one C-like Ig domain, a transmembrane region, and a short cytoplasmic domain. PD-L2 is expressed on antigen presenting cells, placental endothelium and medullary thymic epithelial cells, and can be induced by LPS in B cells, INF-γ in monocytes, or LPS plus IFN-γ in dendritic cells. The CD28 and B7 protein families are critical regulators of immune responses. PD-L2 and PD-L1 are two ligands for PD-1, member of the CD28/CTLA4 family expressed on activated lymphoid cells, and thus provide signals for regulating T cell activation and immune tolerance. The interaction of B7-DC/PD-1 exhibited a 2-6-fold higher affinity compared with the interaction of B7-H1/PD-1.
Programmed cell death 1 ligand 2 (PD-L2), also known as butyrophilin B7-DC or PDCD1 ligand 2, belongs to the member of B7 family which can regulate the activation and tolerance of T cells. PD-L2 is one ligand for Programmed cell death 1(PD-1), and the other is PD-L1. These two ligands shares 34% aa sequence identity. Mouse PD-L2 gene encodes a 273 amino acids (aa) protein with a putative 19 aa signal peptide, a 201 aa extracellular region , a 21 aa transmembrane domain and a 32 aa cytoplasmic region. The mouse PD-L2 gene is highly expressed in heart, placenta, pancreas, lung and liver while expressed weakly in spleen, lymph nodes and thymus. Besides, the expression of PD-L2 gene can be induced on dendritic cells grown from peripheral blood mononuclear cells under CSF2 and IL4/interleukin-4 treatment, and up-regulated by IFNG/IFN-gamma stimulation in monocytes. PD-L2 usually functions in a PDCD1-independent manner and is involved in regulating costimulatory signal which is essential for T-cell proliferation and IFNG production. Recent studies demonstrate that the expression of PD-L2 on the tumor cells promotes CD8 T cell–mediated rejection of tumor cells, at both the induction and effector phase of antitumor immunity. Moreover, PD-L2 binds to PD-1 cells and enhances T cell killing in a PD-1–independent mechanism.
