CD300A is a single-pass type I membrane protein which belongs to the CD300 family. It contains 1 Ig-like V-type (immunoglobulin-like) domain. The CD300 family of myeloid immunoglobulin receptors includes activating (CD300b; CD300e) and inhibitory members (CD300a; CD300f); as well as molecules presenting a negative charge within their transmembrane domain (CD300c; CD300d). It is expressed not only by natural killer (NK) cells but also by T-cell subsets; B-cells; dendritic cells; mast cells; granulocytes and monocytes. CD300A is an inhibitory receptor which may contribute to the down-regulation of cytolytic activity in natural killer (NK) cells; and to the down-regulation of mast cell degranulation. CD300c is a functional immune receptor able to deliver activating signals upon ligation in RBL-2H3 mast cells.

CD300A / IGSF12 is an inhibitory receptor which may contribute to the down-regulation of cytolytic activity in natural killer (NK) cells; and to the down-regulation of mast cell degranulation. CD300c is a functional immune receptor able to deliver activating signals upon ligation in RBL-2H3 mast cells. CD300c signaling is partially mediated by a direct association with the immune receptor tyrosine-based activation motif-bearing adaptor FcεRγ. CD300a and CD300c play an important role in the cross-regulation of TNF-alpha and IFN-alpha secretion from plasmacytoid dendritic cells (pDCs).

CD300LB, also known as CD300b, LMIR5, CLM-7, and IREM-3, is a glycoprotein member of the immunoglobulin superfamily. LMIR5 is expressed on the surface of myeloid lineage cells. It forms noncovalent cis-homodimers and cis-heterodimers with other CD300 family proteins, and the composition of these dimers affects the cellular response. Antibody cross-linking of LMIR5 induces mast cell granule release and cytokine production as well as its tyrosine phosphorylation of LMIR5 (in human). LMIR5 interacts with TIM1 and TIM4 which regulate T cell activation and are themselves binding partners. TIM1 interactions with LMIR5 mediate mast cell activation and the accumulation of neutrophils at sites of TIM1 up regulation on damaged renal tubule epithelial cells. Acts as an activating immune receptor through its interaction with ITAM-bearing adapter TYROBP, and also independently by recruitment of GRB2.
CD30 ligand (CD30L), also known as CD153 and TNFSF8, is a membrane-associated glycoprotein belonging to the TNF superfamily and TNFR superfamily, and is a specific ligand for CD30/TNFRSF8 originally described as a cell surface antigen and a marker for Hodgkin lymphoma and related hematologic malignancies. CD30L is a type-II membrane glycoprotein expressed on activated T cells, stimulated monocyte-macrophages, granulocytes, eosinophils, and some Burkitt-like lymphoma cell lines. CD30L is capable of transducing signals throμgh CD30 on different CD30+ lymphoma cell lines, and mediates pleiotropic biologic effects including cell proliferation, activation, differentiation, as well as cell death by apoptosis. CD30-CD30 ligand interaction has been sμggested to have a pathophysiologic role in malignant lymphomas, particularly Hodgkin disease, large cell anaplastic lymphomas and Burkitt lymphomas, and is also involved in activation and functioning of the T cell-dependent immune response. Thus, CD153 and its receptor CD30 are regarded as therapeutic targets in hematologic malignancies, autoimmune and inflammatory diseases.
The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF8/CD30, which is a cell surface antigen and a marker for Hodgkin lymphoma and related hematologic malignancies. The engagement of this cytokine expressed on B cell surface plays an inhibitory role in modulating Ig class switch. This cytokine was shown to enhance cell proliferation of some lymphoma cell lines, while to induce cell death and reduce cell proliferation of other lymphoma cell lines. The pleiotropic biologic activities of this cytokine on different CD30+ lymphoma cell lines may play a pathophysiologic role in Hodgkin's and some non-Hodgkin's lymphomas. Two transcript variants encoding different isoforms have been found for this gene.
