CD80, also known as B7-1, B7, and BB1, is a 60 kD single chain type I glycoprotein belonging to the immunoglobulin superfamily. CD80 is expressed on activated B and T cells, macrophages, and dendritic cells. CD80 binds to CD28 and CD152 (CTLA-4). Along with CD86, CD80 plays a critical role in regulation of T cell activation. The interaction of CD80 with CD28 provides a potent costimulatory signal for T cell activation through the CD3 complex, while its interaction with CTLA-4 provides an inhibitory signal for T cell activation.
CD81 is a 26 kD non-glycosylated member of the tetraspanin superfamily (TM4SF), also known as TAPA-1 (target of an antiproliferative antibody). CD81 is expressed on T and B cells, NK cells, monocytes, dendritic cells, thymocytes, endothelial cells, and fibroblasts. It also has low levels of expression on granulocytes. CD81 induces B cell adhesion via VLA-4 integrin and has been shown to play a role in early T cell development. CD81 associates with several other cell-surface proteins in a multimolecular complex, including CD19, CD21, CD20, CD37, CD53, and CD82 in B cells, and CD4, CD8 and CD82 in T cells.
CD83 is a 43 kD single chain type I glycoprotein also known as HB15. A member of the immunoglobulin superfamily, CD83 is expressed on a subset of dendritic cells, Langerhans cells, and weakly on activated lymphocytes. Although CD83 is thought to play a role in antigen presentation and/or lymphocyte activation, the precise function of this protein is unknown. CD83 is considered to be a useful marker for mature dendritic cells.
CD86 is an 80 kD immunoglobulin superfamily member also known as B7-2, B70, and Ly-58. CD86 is expressed on activated B and T cells, monocytes/macrophages, dendritic cells, and astrocytes. CD86, along with CD80, is the ligand of CD28 and CD152 (CTLA-4). CD86 is expressed earlier in the immune response than CD80. CD86 has also been shown to be involved in immunoglobulin class-switching and triggering of NK cell-mediated cytotoxicity. CD86 binds to CD28 to transduce costimulatory signals for T cell activation, proliferation, and cytokine production. CD86 can bind to CD152 as well, also known as CTLA-4, to deliver an inhibitory signal to T cells.
